Screening Resorufin Pentyl Ether Analogs for Enhanced Antimicrobial Activity Against Francisella tularensis and Neisseria gonorrhoeae

چکیده
Antibiotic resistance is an urgent public health threat. The CDC estimates there are approximately 2.8 million new cases of antibiotic-resistant infections annually resulting in 35,000 deaths and billions dollars care costs. development drugs imperative to combat this crisis prevent the loss additional lives from once “curable” diseases. Resazomycins, a novel family antibiotics, have bactericidal activity against Francisella tularensis Neisseria gonorrhoeae. One resazomycin, resorufin pentyl ether (RPE), significantly reduces vaginal colonization by N. gonorrhoeae mouse model infection. Repeated administration RPE, however, fails clear infection, contrast single dose ceftriaxone, antibiotic commonly used treat gonorrhea, which clears infection within 24 hours. Further characterization resazomycins revealed efficacy these compounds limited interaction with serum albumin reduced oxygen concentrations found mammalian tissues. Therefore, we hypothesize that resazurin analogs maintain antimicrobial presence low will improved therapeutic vivo. To date, two different derivatives RPE been synthesized tested for F. – 1-methyl 4-methyl RPE. Antimicrobial Resistance and Neisseria gonorrhoeae
Clinical Features N. gonorrhoeae causes infection of mucosal membranes including the cervix, rectum, and throat. If untreated, gonorrhea remains a major cause of pelvic inflammatory disease (PID), tubal infertility, ectopic pregnancy, and chronic pelvic pain. Epidemiologic studies provide strong evidence that gonococcal infections facilitate HIV transmission. Incidence Gonorrhea, caused by Neis...

متن کاملComparative in vitro activity of cefodizime, ceftazidime, aztreonam, and other selected antimicrobial agents against Neisseria gonorrhoeae.
The in vitro activities of three new beta-lactam antimicrobial agents, cefodizime, ceftazidime, and aztreonam (formerly azthreonam), were compared with those of cefotaxime, cefuroxime, cefoxitin, and penicillin against 100 beta-lactamase-negative and 42 beta-lactamase-positive Neisseria gonorrhoeae strains. The three new antimicrobial agents showed excellent activity against N. gonorrhoeae rega...
متن کاملStrengthening Global Surveillance for Antimicrobial Drug–Resistant Neisseria gonorrhoeae through the Enhanced Gonococcal Antimicrobial Surveillance Program
Monitoring trends in antimicrobial drug-resistant Neisseria gonorrhoeae is a critical public health and global health security activity because the number of antimicrobial drugs available to treat gonorrhea effectively is rapidly diminishing. Current global surveillance methods for antimicrobial drug-resistant N. gonorrhoeae have many limitations, especially in countries with the greatest burde...
متن کاملIn-vitro activity of 21 antimicrobial agents against Neisseria gonorrhoeae in Brussels.
The minimum inhibitory concentrations (MIC) of 21 antimicrobial agents was measured for 80 strains of Neisseria gonorrhoeae isolated in Brussels in 1978. Bimodal distributions were found for penicillin G, ampicillin, amoxycillin, carbenicillin, and cephalexin. Of the strains, 17.5% were relatively resistant to penicillin G (MIC greater than 0.08 microgram/ml) 27.5% to ampicillin (MIC greater th...
